A cost effective fermentative production of glutathione by Saccharomyces cerevisiae with cane molasses and glycerol BABT

This work aimed to evaluate the effect of sugar cane molasses and glycerol on glutathione (GSH) fermentation by Saccharomyces cerevisiae ATCC 7754 in flask culture using response surface methodology. Under optimized conditions (80 g/L of molasses and 60 g/L of glycerol), the highest GSH and biomass concentration achieved were 119.6 mg/L and 25.3 g/L, respectively. Further studies done in 5 L bioreactor resulted 241.3 mg/L GSH after 96 h in batch fermentation without amino acids addition and the concentration of biomass was 12.1 g/L. In batch fermentation with the addition of the three amino acids (4 mM cysteine, glycine and glutamic acid at 32 h), biomass reached to 25 g/L and GSH, 236.1 mg/L at 96 h of fermentation. Comparative analyses of response surface methodology and artificial neural networks on incorporating tetracaine into liposomes BJPS

This study evaluated the incorporation of tetracaine into liposomes by RSM (Response Surface Methodology) and ANN (Artificial Neural Networks) based models. RCCD (rotational central composite design) and ANN were performed to optimize the sonication conditions of particles containing 100 % lipid. Laser light scattering was used to perform measure hydrodynamic radius and size distribution of vesicles. The liposomal formulations were analyzed by incorporating the drug into the hydrophilic phase or the lipophilic phase. Extraction of pectin from apple pomace BABT

As apple-processing units are now in expansion in Brazil, industrial by-products like pomace play an important role in pectin manufacture. The objective of this article was to determine a pratical follow-up to the extraction of pectin from apple pomace and to characterize it in a laboratory, on a small scale, aiming at establishing the optimum conditions for acid extraction. The highest yields were obtained when [1]apple pomace was dried and ground to obtain an apple flour to be used as raw material, [2] citric or nitric acids were used and [3] when the citric acid concentration was 6.2 g/100 ml and the time of reaction was 153 minutes. Scale up of dextran production from a mutant of Pediococcus pentosaceus (SPAm) using optimized medium in a bioreactor BABT

The mutant of Pediococcus pentosaceus (SPAm) produced earlier by UV-mutagenesis exhibiting higher dextransucrase activity as compared to wild-type was used. The generated mutant SPAm gave 12.2 mg/ml, a 20% higher dextran than wild-type. Response surface methodology was carried out for further enhancement of dextran production. To enhance dextran production by the mutant SPAm, Plackett-Burman Design and a 2² full factorial Central Composite Design was employed. After response optimization, the optimum concentration of sucrose and yeast extract was 5.115% (w/v) and 0.635% (w/v), respectively. ABSTRACT Clostridial collagenase is recognized as one of the important proteolytic enzymes used in treatment of varieties of fibro-proliferative disorders. The present work aimed to optimise the biomass concentration and collagenase enzyme activity of novel bacilli Clostridium novyi-NT. The response surface methodology tool was used to identify the optimal fermentation parameters. Central composite design (CCD) was applied with respect to three influencing factors - pH, proteose peptone and trypticase soya broth. Statistical optimization of culture conditions by response surface methodology for synthesis of lipase with Enterobacter aerogenes BABT

Optimization of lipase production by Enterobacter aerogenes was carried out using response surface methodology (RSM) where the statistical model was obtained by fractional factorial central composite design. The influence of various physico-chemical parameters, viz. temperature, oil concentration, inoculum volume, pH and incubation period on lipase production was examined. Optimization of physico-chemical parameters resulted 1.4- fold increase in lipase activity. The optimum levels of parameters were 34°C, oil concentration 3%, inoculum volume 7%, pH 7 and incubation time 60 h for obtaining a maximum lipase activity of 27.25 U/ml.
